چکیده

The kinetics and regulation of D-xylose uptake were investigated in the efficient pentose fermentor Candida succiphila, and in Kluyveromyces marxianus, which assimilate but do not ferment pentose sugars. Active high affinity (Km approximately 3.8 mM; Vmax approximately 15 nmol/[mg min]) and putative facilitated diffusion low-affinity (Km approximately 140 mM; Vmax approximately 130 nmol/[mg min]) transport activities were found in C. succiphila grown, respectively, on xylose or glucose. K. marxianus showed facilitated diffusion low-affinity (Km approximately 103 mM; Vmax approximately 190 nmol/[mg min]) transport activity when grown on xylose under microaerobic conditions, and both a low-affinity and an active high-affinity (Km approximately 0.2 mM; Vmax approximately 10 nmol/[mg min]) transport activity when grown on xylose under fully aerobic conditions.

Protein enrichment of an Opuntia ficus-indica cladode hydrolysate by cultivation of Candida utilis and Kluyveromyces marxianus

BACKGROUND The cladodes of Opuntia ficus-indica (prickly pear cactus) have a low protein content; for use as a balanced feed, supplementation with other protein sources is therefore desirable. We investigated protein enrichment by cultivation of the yeasts Candida utilis and Kluyveromyces marxianus in an enzymatic hydrolysate of the cladode biomass. RESULTS Dilute acid pretreatment and enzyma...

Data for rapid ethanol production at elevated temperatures by engineered thermotolerant Kluyveromyces marxianus via the NADP(H)-preferring xylose reductase–xylitol dehydrogenase pathway

Conversion of xylose to ethanol by yeasts is a challenge because of the redox imbalances under oxygen-limited conditions. The thermotolerant yeast Kluyveromyces marxianus grows well with xylose as a carbon source at elevated temperatures, but its xylose fermentation ability is weak. In this study, a combination of the NADPH-preferring xylose reductase (XR) from Neurospora crassa and the NADP(+)...

Production of a Melanin like Pigment by Kluyveromyces marxianus and Streptomyces chibaensis

The aim of the study was to determine the optimum conditions for brown pigment formation by Kluyveromyces marxianus and Streptomyces chibaensis. Pigment formation was followed during the time course experiment in medium contains 20 g/l D-xylose as carbon source, 12 g/l of a mixture of peptone and yeast extract as a nitrogen source at pH 5.0 when incubated at 32°C, the production of pigment reac...

Evolutionary Adaptation of Kluyveromyces marxianus NIRE-K3 for Enhanced Xylose Utilization

The evolutionary adaptation was approached on the thermotolerant yeast Kluyvero myces marxianus NIRE-K3 at 45°C on xylose as a sole source of carbon for enhancement of xylose uptake. After 60 cycles, evolved strain K. marxianus NIRE-K3.1 showed comparatively 3.75and 3.0-fold higher specific growth and xylose uptake rates, respectively, than that of native strain. Moreover, the short lag phase w...

Effect of oxygenation and temperature on glucose-xylose fermentation in Kluyveromyces marxianus CBS712 strain

BACKGROUND The yeast Kluyveromyces marxianus features specific traits that render it attractive for industrial applications. These include production of ethanol which, together with thermotolerance and the ability to grow with a high specific growth rate on a wide range of substrates, could make it an alternative to Saccharomyces cerevisiae as an ethanol producer. However, its ability to co-fer...
